Growth differentiation factor-15 and cardiovascular dysfunction and disease: malefactor or innocent bystander?
نویسندگان
چکیده
It is widely recognized that around 20% of cardiovascular events occurring in the general population cannot be inferred from conventional risk factors. This led to a growing interest in novel measurable humoral biomarkers, hopefully leading to an improvement of global cardiovascular risk stratification. Growth differentiation factor-15 (GDF-15) is the latest promising marker in this field. Growth differentiation factor-15 is a protein belonging to the transforming growth factor-b (TGF-b) superfamily, which includes more than 40 proteins with crucial functions in adult tissue homeostasis by modulating cell survival, proliferation, differentiation, and tissue repair in different organs. Initially cloned from an activated macrophage cell line, GDF-15 has been proposed as a cell death promoter in several tumour cell lines, highlighting its potential role as an inductor of apoptosis in some cancer cells. Expression of GDF-15 is rapidly induced by IL-1, TNF-a, and TGF-b in macrophages, thereby limiting macrophage activation and inflammation. Results of analysis from human tissues evidenced the presence of GDF-15 also in the kidney, prostate and placenta, breast and colon, as well as in the heart.

Commentary on: Is uric acid an indicator of metabolic syndrome in the first-degree relatives of patients with type 2 diabetes?
| March 2013 | 267 other and to uric acid levels. It should be noted that, uric acid is usually considered as an “innocent bystander” of deranged hemodynamic and metabolic status and not as the initiator. However, recent studies showed that elevated uric acid levels are a significant determinant for future HTN, kidney disease, cardiovascular disorders, and also mortality.[1,10,11] These finding...
متن کاملFGF23 from bench to bedside.
There is a strong association between elevated circulating fibroblast growth factor-23 (FGF23) levels and adverse outcomes in patients with chronic kidney disease (CKD) of all stages. Initially discovered as a regulator of phosphate and vitamin D homeostasis, FGF23 has now been implicated in several pathophysiological mechanisms that may negatively impact the cardiovascular and renal systems. F...
متن کاملRadiation Bystander Effects Mechanism
Introduction: Radiation Induced Bystander Effect (RIBE) which cause radiation effects in non-irradiated cells, has challenged the principle according to which radiation traversal through the nucleus of a cell is necessary for producing biological responses. What is the mechanism of this phenomenon? To have a better understanding of this rather ambiguous concept substantial number of original ...
متن کاملThe effect of aerobic training on serum levels of Growth differentiation factor-15 and insulin resistance in elderly women with metabolic syndrome
Background and Objective: Aerobic training plays an important role in the improvement of cardiovascular-metabolic disorders associated with aging and metabolic syndrome; however, the role of this type of training on growth differentiation factor-15 (GDF-15) as a new biomarker for predicting cardiac disorders is not understood. Therefore, the purpose of the present study was to investigate the e...
